چکیده

Introduction Oestrogens are able to regulate gene expression by binding to specific receptors that function as transcription factors [ 1-31. Hormone binding is required initially for the dissociation of an inactive, oligomeric, receptor complex to allow receptor dimerization and high-affinity DNA binding [4, 51 and, secondly, to produce the full transcriptional activity of the receptor [6-91. In view of the role of oestrogens as mitogens in certain types of cells and the observation that approximately 30% of breast cancer can be treated with endocrine therapy, a large number of anti-oestrogens have been developed as potential drugs. T o date these antioestrogens appear to fall into two groups. One group, which includes the non-steroidal antioestrogen tamoxifen, also act as partial agonists while another group, which includes ICI 164384, appear to act predominantly as ‘pure’ anti-oestrogens [ 10, 1 11. The molecular basis for the action of different ligands is now beginning to emerge and is the subject of this review.

Phytoestrogens by inhibiting the non-classical oestrogen receptor, overcome the adverse effect of bisphenol A on hFOB 1.19 cells

Objective(s): Since bisphenol A (BPA) induces bone loss and phytoestrogens enhance the osteoblastogenesis by binding to the non-classical and classical oestrogen receptors, respectively, the present study was aimed to observe the osteoprotective effect of phytoestrogens on BPA-induced osteoblasts in hFOB 1.19 cells.Materials and Methods:...

Oestrogen receptor b ligand: a novel treatment to enhance endogenous functional remyelination

Demyelinating diseases, such as multiple sclerosis, are characterized by inflammatory demyelination and neurodegeneration of the central nervous system. Therapeutic strategies that induce effective neuroprotection and enhance intrinsic repair mechanisms are central goals for future therapy of multiple sclerosis. Oestrogens and oestrogen receptor ligands are promising treatments to prevent multi...

A modified oestrogen receptor ligand-binding domain as an improved switch for the regulation of heterologous proteins.

A number of proteins have been rendered functionally oestrogen-dependent by fusion with the hormone-binding domain of the oestrogen receptor. There are, however, several significant disadvantages with such fusion proteins. First, their use in cells in vitro requires phenol red-free medium and laborious stripping of steroid hormones from serum in order to avoid constitutive activation. Secondly,...

An evolvable oestrogen receptor activity sensor: development of a modular system for integrating multiple genes into the yeast genome.

To study a gene interaction network, we developed a gene-targeting strategy that allows efficient and stable genomic integration of multiple genetic constructs at distinct target loci in the yeast genome. This gene-targeting strategy uses a modular plasmid with a recyclable selectable marker and a multiple cloning site into which the gene of interest is cloned, flanked by two long regions of ho...
